1-ionotrophic-affect permeability of ion channels
2-interact with JAK kinases-affect protein synthesis
3-affect enzymatic activity- affect proliferation/differentiation
4-G-protein receptors and second messenger model
intracellular receptors-cytosolic
are lipid soluble so can directly diffuse into nucleus and affect transcription of DNA so protein synthesis.
can bind to guanylyl cyclase and activate CGMP as second messenger
Camp
1-1st messenger binds to receptor on C.S.M.
2-this triggers sub-unit if G-protein to diffuse across plasma membrane and bind to adenyl cyclase
3-adenyl cyclase cause Camp to be activated
4-this activates protein kinases which phosphorylates proteins
phospholipase C
same principle.
Phospholipase C activates IP3 and DAG.
IP3 acts on ER and releases Ca2+ from internal storage
DAG acts on protein kinases.
main functions of receptors?
1- enable specificity
2- allow appropriate response- membrane permeability, proliferation and differentiation, metabolism, contractile activity, secretory activity.
where does intracellular Ca2+ come from?
1- can come from IP3 release,
2- from outside of cell-ligand-gated channels
3-inhibition of transport out of Ca2+
