Aim
Determine the time taken for starch to be digested and to calculate the rate of enzyme activity at different pH levels
Step one
Add 2 cm³ of each solution ranging from pH 3 to 7 into each labelled test tube using a syringe
Step two
Immerse test tube, starch and Amylase solution in water bath at 25°C
Step three
Place 2 cm³ of Aminé solution into test tube of buffer solution
Step four
Place 2 cm³ of starch into test tube and start time
Step five
Repeat step for every 30 seconds until the iodine solution remains brown and does not turn blue black
Step six
Calculate the rate of enzyme action using:
1 / Time taken for iodine solution to remain brown
Step seven
Repeat steps 1–6 for different Ph values
