Enzyme kinetics 5 Flashcards
1
Q
Where is the major site of drug metabolism?
A
The liver
2
Q
Why do we metabolism drugs?
A
To make them less active
3
Q
What does ADME stand for?
A
Adsorption, distribution, metabolism and excretion
4
Q
What is phase 1 metabolism?
A
Involves the introduction or unmasking of a functional group e.g. by oxidation or hydrolysis (OH, -SH, -NH2, -COOH, etc). Increased polarity of products hence increased water solubility and excretion in urine.
5
Q
Give example of oxidation enzymes used in phase 1 metabolism?
A
- Cytochrome P450 monooxygenase system
- Alcohol dehydrogenase
- Aldehyde dehydrogenase
- Flavin-containing monooxygenase system
- Monoamine oxidase
6
Q
Give examples of reduction enzymes used
A
- NADPH-cytochrome P450 reductase
- Reduced (ferrous) cytochrome P450
