What are 5 sources of dsDNA breaks?
A
immune system rearrangements, meiosis, single-strand nick encountered during replication, insertion by retrovirus & transposon, ionizing radiation, medical imaging

Differentiate between NHEJ & HR
A
NHEJ: required for ionizing resistance & VDJ rearrangements. It simply brings together the 2 ends. Lose a few nts but its quick and is sequence independent.
HR: Occurs in S/G2, needs sister chromatid, but perfectly repairs section. 5’ end is chewed away leaving 3’ ss. It invades homolgous DNA –> holiday junction, later resolves. In meiosis, this is how crossover happens.

Why would misrepair here lead to cancer?

Misrepair –> genomic instability.
