MoA
Antagonises D2 receptors in chemoreceptor trigger zone.
Acts directly on gut to promote gastric emptying; prokinetic effect
MHRA
Risk of cardiac SE
- QT prolongation
- Ventricular arrhythmias
- Sudden death
Counsel patients to report signs of arrhythmias (syncope, palpitations)
Restrictions
12+
Over 35 kg
- Children under 35 kg = 250 mcg/kg TDS)
1 week
10 mg TDS
When is domperidone the choice antiemetic?
Parkinson’s disease as it does not cause extrapyramidal SE
Indications
Symptomatic relief of N + V
Choice antiemetic in PD
Contraindications
Impaired cardiac conduction
Cardiac disease
Severe liver impairment
Interactions
Enzyme inhibitors
- Amiodarone
- Ketoconazole
- Erythromycin
