DNA Replication Flashcards
What are the 5 steps of DNA replication?
1) The original DNA molecule
2) Enzyme DNA helicase breaks the hydrogen bonds between the 2 DNA strands, causing them to separate into 2 single strands
3) Free nucleotides that have been released from the separating of the 2 strands are attracted to their complementary bases along the 2 DNA strands. (A & T / C & G)
4) Once the activated nucleotides are lined up, they are bonded together by the enzyme DNA polymerase to form the sugar phosphate backbone. Hydrogen bonds also form between the 2 complementary bases.
5) All the nucleotides are joined to form a completely new polynucleotide chain. The 2 strands are identical replications of the parent DNA, with one strand being the original template strand and the other being the newly formed strand= semi conservative replication
Following the semi conservative replication theory, describe where a band on nitrogen would form over 3 generations of DNA replication if nitrogen 15 was added to nitrogen 14.
Generation 1: Band forms in the middle= equal strands of nitrogen 15 and nitrogen 14
Generation 2: 2 bands form, thicker one at top (N14) and one in middle (N15)
Generation 3: 2 bands form, much thicker one at top (N14) and one in middle (N15)
