Metformin
Increase peripheral insulin sensitivity and hepatic glucose uptake.
Lactic acidosis and GI disturbance.
Sulfonyureas
Depolarise islet cells in the pancreas increasing insulin release.
Hypoglycaemia and weight gain.
Thiazolidinediones
Activate PPAR-gamma receptor in adipocytes to promote adipogenesis and fatty acid uptake
Fluid retention, weight gain and worsening heart failure.
SGLT2 Inhibitors
Increase urinary glucose loss.
Diabetic Ketoacidosis when used with insulin and increased risk of urinary tract infections.
DPP4-Inhibitors
Inhibit GLP1 breakdown.
Hypoglycaemia and GI upset.
GLP1 Analogues
e.g. Exenatide
Incretin mimetic which inhibits glucagon secretion
Hypoglycaemia, GI upset and may increase the risk of pancreatitis when used with DPP4-inhibitors.
Intestinal Alpha-Glucosidase Inhibitors
e.g. Acarbose
Delay intestinal carbohydrate absorption
Flatulence and GI disturbance.
