Definitions Flashcards
Define pharmacokinetics
Balance between accumulation and elimination
Factors that infuence tissue drug concentration
Absorption, Distribution, Metabolism, Elimination
Define pharmacodynamics
Effect of drug on human body
Pharmacogenetics
Genetic factors affecting drug metabolism e.g. polymorphisms
Absorption
Transfer of drug from site of administration to blood stream
Mechanisms of drug absorption
Active (against gradient, need energy, often need enzyme help) and Passive (with gradient, not needing energy)
By what age are active and passive methods of absorption are fully matured?
4 months
Factors affecting oral absorption include
Gastric acid secretion, gastric emptying time, bile salt foemation, intestinal motility, microbial flora
Example of lipophillic drug
Diazepam
What is first pass metabolism
Pre systemic metabolism i.e. extent to which oral drug is reduced after it’s first passage through an organ prior to reaching systemic circulation (liver, lungs, vascular endothelium)
