Cultivation Flashcards
facultative anaerobes
O2 is needed, but can ferment when must
aerotolerant anaerobes
fermentation, but can tolerate the presence of O2
microaerophils
(needs decreased O2, and increased CO2 concentration
capnophils
(needs increased CO2 concentration
Different Methods to do Anaerobic culturing.
anaerostat (closed chamber, filled with N2-CO2 mixture)
* Gas-Pack jar (closed chamber, sodium-bicarbonate salts produce CO2 and water)
* anaerobic incubator (sealed thermostat filled with CO2)
* certain liquid media (O2 is removed via boiling before culturing, inoculation and
sealing)
* adding chemical substances may help (thioglycolate)
Generation time for colonies is
20mins
colonies visible time is
24 hours
tell ?me abt semi solid media in clturing of bacteria
0,7% agar concentration)
enrichment media?
media used to allow faster growth of certain species than others due to special nutrient content
special media?
- media to culture particular pathogens only
selective media
only certain bacteria are able to grow due to antibiotic or dye supplementation
BOUILLON WITH MEAT method is used for cultivation of which bacteria ?
Clostridia
breaks down the meat
produces gas
(paraffin seal is pushed up is done by which bacteria?
Clostridium perfringes
does not break down the meat
clostridium tetani
