CP5-Beetroot Membrane Flashcards
1
Q
Objectives- investigate the effect of temperature on membrane permeability
A
Objectives:
-know how to effect of temperature on membranes can be determined
2
Q
Risks?
A
- water baths/Bunsen burners can burn you
- cork borer and knives are sharp
- be careful around neat sources, remove anything flammable, keep on safety flame when not in use
- be careful when using sharp items and cut away from self
3
Q
Method?
A
- prepare 8 water baths with temperatures ranging between 0 and 70 degrees, at 10 degree intervals, using a thermometer
- use a syringe to add 10 cm^3 of distilled water to 8 test tubes and label with temperatures from the range of water baths
- place each tube in a water bath for 10 minutes
- cut 8 pieces of beetroot using the same cork borer and cut to the same length of 1cm long and wash the cylinders until they run clear and dry gently with a paper towel
- add one beetroot to each water bath and leave for 15 minutes
- set the colorimeter to 0 by using a cuvette of distilled water as a reference value
- pour 2cm^3 of the supernatant from each water bath into a cuvette and record the absorbance of the solution without the beetroot piece
- record in a table and plot a graph of transmission against temperature
4
Q
Variables?
A
IV-the temperature of the water bath
DV-the absorbance of the supernatant
CV- size and mass of beetroot, type of beetroot, same pre-treatment
