Why can liver disease lead to an inability to coagulate?
- Many Coagulation Proteins are made in the Liver
What must happen for a zymogen to get activated?
- Must have protease cleave a portion to convert the -ogen to an active protein
What happens has a result of cleaving the following zymogens?
- X
- Fibrinogen
X —> Xa, protein is cleaved and activated to give it PROTEASE activity
Fibrinogen —> Fibrin, protein is cleaved allowing it to form AGGREGATES
What does fibrinogen do when it gets bound to Xa?
Accelerates coagulation by 1000x
What happens when proteins (factor VII receptor) on non-vascular cells are exposed to the blood?
Proteins on their surface trigger initiation of coagulation response
How are downstream clots prevented in the clotting response?
- Anti-coagulation Proteins displayed by endothelial cells bind other Anti-coagulation proteins and destroy Accelerators of coagulation
What is the structural Protein of a clot?
Fibrinogen/Fibrin
What is the purpose of fibrin crosslinking in the clotting response?
- Makes the Clot strong enough to resist hemodynamic and other forces
