Q

Why must the starting line be drawn with a pencil not a pen

A

Dyes from the pen ink will result in additional separated components introduced into the chromatogram

Q

How to identify colour less substances (for example amino acids)

A

Spray/add locating agents such as (ninhydrin) onto the chromatogram, which reacts with (amino acids) to form coloured products, thus making them visible

Q

whats retention factor, how do u calculate it

A

divide the distance travelled by the component by the distance travelled by the solvent. (the answer is always less than 1)
lets say the solvent travelled 6cm and the component only travelled 3cm so 3/6 = 0.50 retention factor

Q

when doing 2d chromatography, y does the first one have less spots

A

this is because they have the same solubility in solvent x and will have the same retention factor
