- ALONE: 100mg/m2 q21d (D1, 22, 43)
- WITH 5FU: 20mg/m2 D1-4 & D22-25 w/ 1000mg/m2 5-FU
- Myelosuppression (anemia, thrombocytopenia, neutropenia), nephrotoxic, peripheral neuropathy,
ototoxicity, N/V, mucositis - Carboplatin: Decreased oto/neuro/nephrotoxicity; myelosuppression main S/E - 15-30% single agent partial response

Q: Mechanism of action & S/E of MTX [?]
A
- = Anti metabolite
- Interferes with cellular metabolism in S phase of cell cycle via inhibiting dihydrofolate reductase enzyme (required for folate → tetrahydrofolate in S phase)
- Causes hepatic fibrosis
- Rescue treatment → folinic acid (leuocovorin)

Q: Mechanism of action & S/E of 5-Flurouracil (5-FU) [?]
A
- = Anti metabolite
- Interferes with cellular metabolism in S phase by (i) creating Uracil analog in S phase that prevents DNS replication OR (ii) inhibition of thymidylate synthetase .
- S/E: Cardiotoxicity, mucositis, dermatitis, myelosuppression
