Methotrexate

A

An antimetabolite (Compete with purine or pyrimidine analogs by getting incorporated into RNA/DNA inhibiting synthesis, targets S phase of cell cycle)

Folate antagonist

Indications: ALL, breast ca, head and neck ca, non-Hodgkin’s lymphoma, osteosarcoma

Adverse Effects: Acute N,V, mucositis,
Delayed: myelosuppression, nephrotoxicity, hepatotoxicity, photosensitivity, pulmonary toxicity

A

Indications: Breast, colon and gastric CA

Adverse Effects - Acute: N/V/D
Delayed: Stomatitis, GI ulcer, hand-foot syndrome, cardiotoxicity, photosensitivity, myelosuppresion

topoisomerase inhibitors

A

Etoposide, Irinotecan

MOA: Binds topo I and stabilizes cleavable complex, so relegation of cleaved DNA strand cannot occur. Leads to DNA strand break. Targets S phase of cell cycle.

Indications: small cell lung ca, NHL
