ch15: sequencing Flashcards
DNA sequencing
determination of actual base sequence of DNA
dideoxynucleotides (ddNTPs) are used as
chain terminators in DNA synthesis reactions; cannot code for anymore bases once attached
gel electrophoresis
separates DNA strands by size
genes are identified by
open reading frames
sequence annotation
the addition of information, such as ORFs, to the basic sequence information
plasmids
small circular chromosomes that replicate themselves because they have an origin of replication
three parts of a plasmid
1) origin or replication
2) selectable makrer
3) multiple cloning site
SNPs (single nucleotide polymorphisms)
single nucleotide base substitutions that occur in more than one percent of the general population; about one in every 300 nucleotide base pairs
haplotype
combination of SNPs used to identify groups of individuals with similar DNA
