Adoptive transfer of Pmel-1 T cells along with a vaccine encoding the altered peptide ligand and administration of exogenous IL-2 –> regression of established B16 melanoma; T cell infiltration and proliferation are antigen independent but tumor killing is antigen dependent.
Confirm transgenes by PCR analysis for the pmel-1 TCR a and b chains.
Mice injected i.v. with 2x107 PFU of recombinant flow pox encoding human gp100 and injected i.p. with 6x106 IU of recombinant human IL-2 in PBS, twice daily for 5/6 doses
The presence of B16 tumor alone did not activate trancgenic cells; no T cells found in unmanipulated pmel-1 transgenic mice but found in the tissue in large numbers after administration of IL-2 and vaccination.
