ABO Typing
Forward: add one drop of anti serum and patient cells
Reverse: add two drops of patient serum and one of reagent cells
Centrifuge, grade reactions
ABO Discrepancies
Aquired B: an unexpected B antigen is detected on the reverse test, check history for colon issues
Mixed Field: 4+ agglutination with a cloudy background, indicates a recent transfusion
Weak Antigen: expected antigen has a weak or absent rxn, incubate for 15 min and test again, caused by old age and immunocompromised
Rh Typing
Add one drop of anti serum and patient cells
Incubate and record the rxn
Any rxn means Rh positive
Weak D Procedure
Do for babies of Rh-negative mothers and donor typing
After IS, incubate at 37C for 15 min
Wash x4, add AHG and read
D con Procedure
Do for AB+ patients
Add two drops of monoclonal reagent and one drop patient cells
Antibody Screen
Add two drops of patient serum to each tube
Add one drop of cell I, cell II, and patient cell to the corresponding tubes
Instant spin
37C incubation
Wash x4 and add AHG
Check all negative tubes under the microscope and add check cells
Antibody ID
Add two drops of patient serum and one of corresponding test cells to each tube
IS, 37C, AHG phases
Check all negative tubes under the microscope and add check cells
Post Natal Testing
ABO and Rh type mom and baby
Antibody screen mom
If mom is Rh-, baby Rh+, do DAT on the baby
If DAT or antibody screen positive, do FMHS on the mom

Crossmatching
Get three tubes with patient ID, donor ID, and initials on them
Get the donor section, add two drops to one tube to make a red cell suspension
Put the section in a labeled tube and cover
Add two drops patient serum and one drop donor cells to XM tube
IS if no patient issues
IS, 37C, AHG phases if there are patient antibodies
