bio microscope test Flashcards
how do you carry a microscope
in an up right position, one hand under the base, and the other hand on the arm
what are the 5 steps to putting away a microscope
1- turn it off
2- the stage is all the way up
3- place on the scanning objective (the red one and the smallest)
4- eye piece even with the arm
5- clean microscope with an alcohol wipe but not the lenses
how do you clean a microscope slide
you can only use lens paper while cleaning it
what are the steps to making a temporary slide
1- put the object on glass slide and make sure its small enough to fit under the cover slip
2- drop water on the object
3- with forceps (tweezers) place the cover slip over the object
(if there are large water bubbles drop water right next to the cover slip)
how do you stain a slide
1- make a temporary slide and the cover slip MUST be flat
2- drop lodine on one side of the cover slip
3- use the paper towel square on the other side to pull it through
(you can clean this uses lens paper)
how do you find an object on the low power objective
1- adjust the diaphragm
2- bring the stage all the way up
3- move the course adjustment slowly till the object comes into focus
4- use the fine adjustment to bring out detail and make it cleaner
how do you fins an object on the high power objective
1- find it on low power first
2- without moving the stage turn to high power
3- center the object
4- use the fine adjustment only to bring out the detail
never use the course adjustment
identify all parts of the microscope
look at notes (no word bank)
explain the characteristics of a compound light microscope (a-e)
A- use light to form on image
B- provides a flat image
C- can be used on living and non-living things
D- the higher the magnification, the worse the resolution
E- highest magnification is 2000x
What are the advantages and disadvantages of a compound light microscope
advantages - can be used on living and non-living things
disadvantages - the higher the magnification, the worse the resolution
what type of microscope is used in class
compound light microscope
define resolution
the clarity or sharpness of the image
define image
what you see under the microscope
define iris diaphragm
has a central opening with over lapping plates that can be adjusted to increase or decrease the control opening
define disc diaphragm
consists of a flat plate, with various sized holes
define ocular lens
another name for the eye peice
