BCHM drugs Flashcards
Azidothymidine (Zidovudine or AZT)
Structure?
Anti-viral Nucleoside Analogue of deoxythymidine
No 3’ OH Group due to N=N=N group at 3’ position
Azidothymidine (Zidovudine or AZT)
Target?
Viral reverse transcriptase
Inhibitor of viral replication
Acyclovir
Structure?
Anti-viral Nucleoside Analogue of deoxyguanosine
No 3’ OH Group due to broken ring structure of pentose sugar
What is special about Acyclovir
First phosphorylation by viral thymidine kinase (specificity b/c only infected cells possess viral kinase), last two phosphorylations by host cellular kinase. Causes chain termination once incorporated to DNA.
What is the target of Didanosine?
Viral reverse transcriptase
Inhibitor of viral replication
HIV
Adenosine arabinoside (Vidarabine) used for which disease?
Relapsed Childhood Acute Lymphoblastic Leukemia and Herpes Simplex Infections
Which drugs can be used for HIV?
Didanosine
Tenofovir
AZT
How does 5-aza-2’-deoxycytidine (Decitabine) – 5-azacytidine work?
Inhibits Methylation once incorporated into DNA and RNA
Which drug do you use for TB?
Rifamycin (Rifampin)
In cells, which drug is converted to FdUMP which inhibits thymidylate kinase, and thus inhibiting synthesis of thymine nucleotides that are required for DNA synthesis.
5-fluorouracil
Which drugs attach viral DNA polymerase?
Tenofovir and Adenosine arabinoside (Vidarabine)
Inhibits HUMAN DNA polymerase
Cytosine arabinoside (araC or Cytarabine)
Inhibits DNA Gyrase
Novobiocin, Ciprofloxacin and Nalidixic Acid
Which drug intercalates between adjacent G-C Bases
Actinomycin D (Dactinomycin)
What does Alpha-Amanitin Toxin inhibit?
RNA Polymerase II (Eukaryotes)
Which drug binds to 30s ribosomal subunit of prokaryotes and distorts structure, preventing initiation of translation?
Streptomycin
