Batch and Continuous culture Flashcards
1
Q
What is on the y axis on a batch culture graph?
A
- log number of microorganism
2
Q
what is on the x axis on a batch graph?
A
- Time
3
Q
What is happening in the lag phase?
A
- genes activated e.g. lactose permease
- enzymes transcribed and translated
- substrate is being broken down
- slow population growth
4
Q
What is happening in the log/Exponential phase?
A
- Rapid population growth
-abundant nutrients - cell production greater than cell death
5
Q
What is happening in the stationary phrase?
A
- cell production = cell death
- lack of nutrients/build up of waste products
- carrying capacity reduced
6
Q
what is happening in the death phase?
A
- death rate greater than cell population
lack of nutrients/build up of waste products - expression of anti-microbial compounds
- e.g. penicillium makes penicillian
7
Q
What is the batch culture method?
A
- closed fermentation vessels
- microorganisms and nutrients added at start
- nothing added or removed
products are separated at end - temperature is usually controlled e.g. using water jacket
- paddles/stirrers used to stir
- even distribution of nutrients
- nutrients are depleted at the end
8
Q
What is continuous culture method?
A
- open fermentation vessel
- nutrients added and products removed at a constant rate
- maintains microbes in exponential phase
- conditions kept constant
e.g. temperature, PH, oxygen, glucose
