What is AUC used for?
Help determine the quaternary structure by studying the behaviour of macromolecules in solution under the influence of a strong gravitational force
- macromolecules either sink or float
What is centrifugation?
- separation achieved by exerting a force by spinning
- force exerted is governed by distance from the axis and (speed rotation)^2
What is the fastest shape for sedimentation?
A sphere
What observations can be made using AUC?
dynamic behaviour (sedimentation velocity) or of systems in equilibrium (sedimentation equilibrium) - provide info about size, shape, density and conformational changes in proteins and other macromolecules
What does the sedimentation velocity experiments show?
Sedimentation velocity experiments aim to interpret the entire time-course of sedimentation, and report on the shape and molar mass of the dissolved macromolecules, as well as their size-distribution
How does the sedimentation velocity show molar mass?
Using the Svedberg Equation
