What are the properties of the Arterivirus virions and genome?

A

Enveloped, spherical, 60nm diameter. Isometric nucleocapsids with small glycoprotein spikes. The genome is linear, positive sense, ss RNA, 12kb size with 9-12 ORF.

What is the replication strategy of Arteriviruses? Where does it take place?

A

They attach using the env protein, and enter through low pH dependent endocytic pathway. Replication and transcription is done through a full length neg strand intermediate, from which subgenomic negative strands of mRNAs are transcribed. 2 large ORF at 5’ end with repicase proteins are modified into 12 nsp that mediate replication. The negative strand RNA template is transcribed to positive strand subgenomic RNAs that are translated into individual virion proteins. This happens in cytoplasm. Newly synthesized nsp are incorporated into RER as double membrane vesicles. New genome is encapsidated with N prot, enveloped by budding our of RER with viral env prot, released by exocytosis.
