A

Class: Antitumor abx

MOA: Induces free radical formation
–> breaks in DNA strands

Clinical use: Testicular cancer, Hodgkin
lymphoma

Adverse effects: Pulmonary fibrosis, skin
hyperpigmentation. Minimal
myelosuppression

Dactinomycin

actinomycin D

A

Class: Antitumor abx

MOA: Intercalates into DNA,
preventing RNA synthesis

Clinical use: Wilms tumor, Ewing sarcoma,
rhabdomyosarcoma. Used for
childhood tumors

Adverse effects: Myelosuppression.

Doxorubicin,

daunorubicin

A

Class: Antitumor abx

MOA: Generate free radicals.
Intercalate in DNA --> breaks in
DNA --> ↓ replication.
Interferes with topoisomerase II
enzyme

Clinical use: Solid tumors, leukemias,
lymphomas

Adverse effects: Cardiotoxicity (dilated
cardiomyopathy),
myelosuppression, alopecia.
Dexrazoxane (iron chelating
agent) used to prevent
cardiotoxicity
