Antiparkinsonian drugs Flashcards
what is parkinsons disease
A progressive neurodegenerative disorder characterized by bradykinesia, rigidity and tremors
What is parkinsons caused by
loss of dopaminergic neurons due to degeneration of substantia nigra
What drugs can cause parkisonism effects
Neuroleptics-typical like haloperidl, droperidol clomprazine
What is the aim of antiparkinson drigs
increase dopamine acitivity
decrease cholinergic activity
Main dopamine therapy
levodopa and carbidopa (dopamine decarboxylase inhibitor)
Carbidopa prevents peripheral synthesis of dop from LDOPA
So more levodopa available to CNS
Why is L-DOPA used not dopamine
because it can cross blood brain barrier
Monoamine Oxidase B inhibitors
Selegiline, Raselegiline
prevents dopamine metabolism
Do MAO B inhibitors cause cheese syndrome
No
MAO A inhibitors for antidepressents causes cheese syndrome bc prevents breakdown of tyramine - sympatic mimetic activity
COMT inhibitors
Entacapone, Tolcapone
Blocks enzyme that breaks down LDOPA
Dopamine receptor agonists
Bromocriptine, Ropinirole
Which drugs are dopamine recpetor ANTAGONISTS
neuroleptics
haloperidol, chlorpramizine, clozapine
Glutamatergic blockers
Amatadine
Amantadine is also used in what
has anti INFLUENZA A activity
Cholinolytics
atropine, biperidin
atropine not used bc of side effects
WHat is Crura Bulgara
the first therapy for parkinsosn diesease dveloped by a bulgarian
made from Atropa Belladonna
Adverse eefects
Peripheral- nausea vomit tachycardia hypotension
CNS- mood changes depression psychosis audotiry hallucinations
