Benzimidazoles - indications
nematodes (ascariasis, lymphatic filariasis - combo)
Lymphatic filariasis - treatment
Albendazole + ivermectin or diethylcarbamazine (1/yr x 5 yr)
Benzimidazoles - mechanism
inhibits microtubule polymerization (B-tubulin - cochicine)
Also inhibits glucose uptake, mitochondrial fumarate reductase, uncoupling of oxidative phosporylation
Benzimidazole - resistance
B-tubulin isotype 2, or type 1 has phen to tyrosine mute
Mebendazole - drug or prodrug?
Drug
Mebendazole - biologic activity
poor absorption, high 1st pass
Mebendazole - indications
GI tract infections (little systemic)
Albendazole - prodrug or drug?
Prodrug (converted to albendazole sulfoxide)
Albendazole - administration
PO, fatty food enhances
Albendazole - penetration
tissues and hydatid cysts
Albendazole - indications
