Anti-virals Flashcards
What is the mechanism of action of acyclovir?
Acyclovir is a acyclic guanine nucleoside analogue that once phosphorylated, stops replication of viral DNA in 3 ways:
- Competitively inhibits viral DNA polymerase
- Inactivates viral DNA polymerase
- Is incorporated into the viral DNA chain, preventing the elongation of the viral DNA chain
Acyclovir is the synthetic analog of which nucleoside?
Guanine
What is the difference between acyclovir and valacyclovir?
Valacyclovir is the prodrug of acyclovir; it has much better oral bioavailability
What is acyclovir indicated in?
HSV1, HSV2, VZV & EBV
Acyclovir & valacyclovir is contraindicated in which group of individuals?
Patients with renal impairment
What is the mechanism of action of ganciclovir?
Ganciclovir is a acyclic guanine nucleoside analogue that once phosphorylated, stops replication of viral DNA in 3 ways:
- Preferentially inhibits viral DNA polymerase
- Inactivates viral DNA polymerase
- Is incorporated into the viral DNA chain, preventing the elongation of the viral DNA chain
What is ganciclovir indicated in?
All herpesviruses especially CMV
How is ganciclovir administered?
IV; prodrug valganciclovir has better oral bioavailability
How is acyclovir administered?
PO/IV/topical
What is the main adverse effect of acyclovir?
Nephrotoxicity
