Give 5 rules of BSL4 work

A

2 person rule.
Complete change of clothes before entering.
Positive pressure suits with separate air supply.
Shower on exit.
Strict waste and material management.

How is waste handling different in BSL3?

A

Solid waste has two autoclave bags in the hood, then the steel container has an extra one.
Liquid waste contains more concentrated bleach at 13%, and a vacuum is used but only for non airborne pathogens.

Could you bring a katana in BSL3 ?

A

Nope. Strict no sharps policy, no glass no razor no needles etc. If absolutely necessary it can be used but have to wear mesh gloves at that moment.

What additional precaution must you take when pipetting aerosol pathogens?

A

Draw up some bleach in the tip before discarding it.

How can risks increase through independant experiments?

A

Well for example if viruses recombine. Therefore, check what others are doing and discuss with BSO and PI what experiments you’re doing in there.

What are the three levels of cleaning in a BSL3 lab?

A

Daily cleans, which is just the regular things we do in the lab.
Intermediate cleans, where every surface, centrifuge is cleaned with ethanol, and floors wiped with Swiffer wipes.
Large cleans, Where you dismantle BSCs and cleaned with stronger disinfectant. Incubators cleaned, and shelves autoclaved. Floors washed with strong disinfectant too. That happens every 6 weeks, in alternance with intermediate cleans.

A spill occurs. Hows do you deal with it step by step?

A

Inform others, evacuate, ask for help if necessary.
Tell BSO. Send anyone to shower.
Put on mask, goggles.
Wait 30 mins (aerosols to settle)
Minimize the spread of the spilll using absorbant stuff.
Put ethanol on top after a while.
Mark contaminated area.
Clean the area, leave for 30 min.
Change top gloves, discard the material in waste.
Inspect the area, wait for permission of BSO, then return to normal.
Refill Spill kit, do a report with BSO.

Something breaks in a centrifuge, What next?

A

Stop the bloody machine.
Warn others and clear area, inform BSO, wait 30 min.
Equip with all the PPE, then open centrifuge. If the contamination is contained in the sealed bucket, open it in the hood under an autoclave bag, then autoclave the whole thing.
Once autoclaved, remove broken vials and dispose of it.

A hood stop working. What do you do?

A

in BSL2, stop pipetting, close all viral vessels, and then anything else. Put a mask on, spray the living sh*t out of the hood inside. Close the hood and tell BSO.
If in BSL3, you start by closing the hood window. put a mask on, inform BSO, evacuate if necessary, then deal with it trying to look if there’s an outage or something else, by calling service center. If it’s working again wait 30 mins. otherwise, leave the lab.

What’s required to ship infectious stuff?

A

A triple layer packaging system. The primary receptacl econtaints the specimen and is watertight, leakproof and labled. It’s wrapped in absorbent material.
Secondary packaging is also watertight etc.
The outer package protects the rest from damage.

Of course, plenty of forms are required for that.
