a1 agonist (a1 > a2)
Indication of Phenylephrine
Nasal (rhinitis) decongestant
MoA of Phenylephrine
Systemically ↑ mean BP via vasoconstriction.
PK/PD of phenylephrine
1) ↑ BP may elicit a reflex bradycardia.
2) Cardiac output may be ↓ but also offset by ↑ venous return.
Adverse effects of Direct-acting Non- Catecholamines
- Anxiety or euphoria.
- Drowsiness or insomnia.
- Hypotension or hypertension (a2 withdrawal)
- Angina.
- Palpitations and arrhythmias
- Cardiac arrest.

Mild/ Moderate hypertension
MoA of Clonidine and Methyldopa
Stimulate prejunctional receptors in CNS to
decrease sympathetic Methyldopa outflow

1) ↑ HR and
2) ↓ mean BP
via vasodilation
