ADME Flashcards
What is the blood brain barrier?
Extreme form of lipid barrier with:
Few intercellular pores and numerous tight junction surrounded by glial cells
Lipid soluble drugs penetrate and water soluble/polar drugs have limited access
What happens to the BBB during meningitis infection?
BBB less effective in meningitis and can give antibiotics which don’t normally pass through - benzylpenicillin
What is the difference between domperidone and metoclopramide?
Both antiemetics
Both dopamine receptor antagonists
But metoclopramide penetrates BBB and can cause drug-induced Parkinsonism unlike domperidone which can’t pass through BBB
Why does a small dose change in phenytoin lead to a disproportional plasma change?
Zero order (non-linear) kinetics of the drug
