Sep-pak QC procedure

A
  1. Sample Preparation (0.1 ml of RP in tuberculin syringe)
  2. Condition Sep-Pak ® (Wet/Activate)
    ▪ 1-2 mL Ethanol, Methanol, HCL, Acetonitrile, Saline or Sterile Water
    (solvent)
  3. Add 0.1 ml of RP sample to the long end (inlet) of the Sep-Pak ®
  4. Rinse Sep-Pak ®
    ▪ Ethanol , Methanol, HCL, saline (depending on RP)
    ▪ Collect each in a separate falcon tube (Fractions) – Bound, Free, HR
    ▪ Push 10 ml of air through the Sep-Pak ® after final rinse
  5. Assay components (Falcon tubes & Sep-Pak ® individually)
  6. Calculate % Radiochemical Purity according to Package Insert

Considerations for sep-pak QC

A
  • accuracy vaires with flow rate and sample load
  • Flow rate: 15-20ml/min
  • FR too high- no interaction
  • Backsplash- contaminations and falsely low RCP
  • RP loaded in wrong end
  • wrong sep-pak used
