8_8_16 Amino Acids and Protein Structure Flashcards
2 main classes of protein
structural and dynamic
Protein structure (primary)
sequence of amino acids going from an amine terminus to a carboxy terminus
Amino Acids (general)
Zwitterionic, pKa of N-term -9, C-term -2, all L configuration (CORN)
potential a.a. modifications
phosphorylation: Ser, Thr, Tyr
O-linked glycosylation: Ser, Thr
N-linked glycosylation: Asn
Disulfide bond: Cys
Henderson-Hasselbach equation
pH = pKa +log([A-]/[HA])
Alpha helix
right handed, dipole, H-bonds (n to n+4), 3.6 residues/turn, 5/4Å/turn, 1.5Å/residue, side chains point out and down at 100˚ intervals
beta-sheets
3 perpendicular dir., 3.5Å/residue, H-bonds between strands, side chains go up and down, antiparallel are non-polar to polar etc.
parallel is all nonpolar
