Competitive inhibitors
has chemical structures resembling substrate
Allosteric sites
-far from active site
-allows allosteric ligand binding at all time (non-competitive)
-allow binging after substrate binds in active site (uncompetitive)
-conformational changes occur after binding, causing enzyme to be less effective
IC50 values
–the conc of inhibitor that causes 50% inhibition of the enzyme
Drugs as a competitive inhibitor
-side effects(interact w/ sim enzymes)
-resistance(target organism mutates enzyme/ degrades or detoxifies drug)
-bioavailability(drug doesn’t reach intended site/degraded/excreted)
Biological feedback
substrate acts as inhibitor, so conc of substrate in cell is not too high/ more than required by cell
