43. Antimalarial drugs Flashcards
Drugs that destroy or prevent development of Plasmodium sp in humans (subset of antiprotozoal drugs)
Treatment for malaria is not initiated until __
diagnosis has been confirmed by lab tests
Antimalarial drugs affect the parasite during its __ cycle
asexual cycle (which takes place in the human body)
Antimalarial class that inhibits DNA and RNA polymerase; disrupting protein synthesis
4-aminoquinolines
Antimalarial class that inhibits an enzyme needed to produce vital substances in malarial parasites
diaminopyrimidines
Antimalarial class that binds to an alters parasitic DNA
8-aminoquinolines
Antimalarial drug contraindications
Drug allergy, tinnitus, pregnancy (quinine), severe renal, hepatic, or hematologic dysfunction
Associated with serious neurologic and psychiatric side effects.
mefloquine
Indicated for treatment of malaria, amebiasis; Anti-inflammatory actions; Contraindications in visual field changes, optic neuritis, and psoriasis; 4-aminoquinolines
chloroquine and hydroxychloroquine
Prevention and treatment of malaria and multidrug-resistant P.falciparum; Black box warning for neuropsychiatric AEs, should not be used in pts w major psychiatric disorders; Analogue of quinine
mefloquine
Can destroy parasites in exoerythrocytic phase; Contraindicated in blood disorders and G6PD deficiency; Similar to 4-aminoquinolones but classified as an 8-aminoquinoline
primaquine
Contraindicated in pts with megaloblastic anemia caused by folate deficiency; diaminopyrimidine
pyrimethamine
