Transmission electron microscope TEMs
Scanning electron microscope SEMs
What are TEMS?
Use electromagnets to focus a beam of electrons which is then transmitted through the specimen.
Denser parts of specimen absorb more electrons, which makes them look darker on the image
Advantages of TEMs
High resolution images
Able to see internal structure of organelles like granum in chloroplasts.
Disadvantages of TEMs
Have to view specimen through vacuum so cannot view living organisms.
Only used on thin specimens.
What are SEMs?
Scan a beam of electrons across the specimen.
This knocks off electrons from specimen which then gather in cathode ray tube to form image.
Advantages of SEMs
Images you end up with show surface.
3D.
Used on thick specimens.
Disadvantages of SEMs
Low resolution images.
Only used on non-living specimens.
