• Inhibit N+/Cl- co-transporter in distal convoluted tubule - ↓Na+ and H2O reabsorption

A
Hypokalaemia, hyponatraemia, hyperuricemia (gout), arrhythmia
↑glucose (especially with beta-blockers)
small ↑cholesterol and triglyceride
