What is the first step?

1. Place one drop of iodine in to each well of spotting tile
2. Put starch solution in a boiling tube
3. Place amylase solution in another boiling tube
4. Add pH buffer to the second boiling tube
5. Place both boiling tubes in a water bath for 2 minutes
6. After two minutes, add the contents of both boiling tubes together
7. After every 30 seconds, use a pipette to put one drop of solution into a new well of the tile
8. Repeat every 30 seconds until it turns blur or black
9. Repeat with different pH buffer solutions
