How does imetelstat work ?
A
- Anti-sense therapy is when a genetic sequence is known to cause a particular disease, then a strand of DNA/RNA can be synthesised that will bind to mRNA produced by the genes and inactivate it.
- Anti-sense oligonucleotides are small RNA/DNA molecules that are used for this purpose.
- E.g.Imetelstat
- Imetelstat is an antisense oligonucleotide that inhibits telomerase activity, and impede cell growth. It is in phase 2 trials for myeloproliferative neoplasms – myelofibrosis and ET

How does sacubril work?
A
- Sacubril is an neprilysin inhibitor
- Neprilysin is responsible for degradation of ANP and BNP, which cause reduction in blood volume through decrease in systemic vascular resistance and increase in natriuresis, and thus decrease in afterload.

What are the benefits of sacubril/valsartan?
A
• Reduction in CVD death, heart failure hospitalistaions, death from any cause by 20% compared to ACE alone

How does riociguat work ?
A
In PAH there is dysfunction of the NO-soluble guanylate cyclase monophosphate pathway. Riocigiuat is an sGC stimulator causing vasodilation,
