what are orphan GPCRs?

A

proteins that have similarities to the receptors whose ligands have not yet been identified

Q

how do MOPRs work in the reward pathway?

A

they disinhibit DA neurons by inhibiting GABA release in the ventral segmental area

Q

What are b-arrestins?

A

scaffolding proteins coupled to GPCRs that can bring signals to outer receptors and is involved in receptor endocytosis leading to tolerance

12
Q

How do B-arrestins lead to endocytosis?

A

phosphorylation of the GPCR by GRK or a BARK complex stimulated by Gbg causes uncoupling of the ligand. recruitment of b-arrestin usually targets the receptor for endocytosis via clatharin coated vesicles

14
Q

What can be observed in a Barrestin2 KO?

A

reduced tolerance, increased basal analgesia, morphine is more rewarding and locomotion slows, respiratory depression, constipation and locomotion are all reduced

16
Q

What are the differences between D1 like and D2 like DA receptors?

A

D1 like - 1,5 - stimulate AC and found in BG and hypothalamus
D2 like-2,3,4 - inhibits AC and found in BG and limbic system

17
Q

What can be observed in TH KO?

A

lack of morphine stimulation of locomotion although can be increased by caffeine - place preference shows there is still a reward for morphine

18
Q

What can be observed in DAR KO?

A

recduced morphine locomotion
b-arrestin is involved
mouse locomotion is dependent on D1 activation of ERK and B-arrestin 2
