What does the enzyme amylase catalyse
The breakdown of starch to maltose
If starch is present, the iodine solution will change from browny orange to
Blue - black
How do you investigate how pH affects amylase activity
1) put a drop of iodine solution into every well of a spotting tile
2) heat water to 35° in a Bunsen burner
3) use a syringe to add 1cm3 of amylase solution and 1cm 3 of buffer solution(pH5) to a boiling tube- put tube into water for 5 mins
4) use a different syringe to add 5cm3 or a starch solution to the boiling tube
5) mix the components and start timer
6) using a dropping pippet, take fresh samples from the boiling tube every 30 seconds and put a drop into each well- when solution remains browny orange starch is no longer present
7) repeat with different pH values
