What is zero-order kinetics?
Constant amount of substance removed per unit time, regardless of concentration
What is volume of distribution? (Pharmakokinetics)
Amount of drug absorbed into the body (dose/plasma)
What factors affect endogenous clearance
Route of exposure, comorbidities, organ dysfunction e.g. toxin causing AKI may have reduced clearance
When is extracorporeal therapy considered in terms of volume of distribution?
<1-2L/kg volume of distribution
Haemodialysis = diffusive clearance, what is the toxin size appropriate? Water-soluble?
Through diasylate. Toxins that are 500-1000 Da, water soluble
Haemofiltration = convection, toxin size? Water-soluble?
Solvent removed, cleared through solvent drag >1000-10,000 Da, better for protein bound toxins
What is contained in intralipid?
20% primarily soybean fat emulsion
MOA of intralipid?
Lipid sink decreasing bioavailability of lipid-soluble medications, delivering FFas to myocardium, stimulate voltage gated Ca channels, increased vasopressor effect (alpha adrenergic)
