Gout Drugs Flashcards
MOA of Indomethacin
inhibits COX, thus inhibiting prostaglandin synthesis and urate crystal phagocytosis
Side Effects of Indomethacin
GI ulceration
bleeding
HA
tinnitus
dizziness
confusion
MOA of Colchicine
Binds to intracellular tubulin, preventing its polymerization into microtubules
- Leukocytes won’t be able to migrate or phagocytize
MOA of Probenecid and Sulfinpyrazone
- completely inhibits the reabsorption of uric acid by renal tubules
- increases the excretion of uric acid Inhibits prostaglandin synthesis
Side effects of Probenecid
Inhibits excretion of other drugs urolithiasis
GI disturbance
Dermatitis
Blood dyscrasias
Side effects of Sulfinpyrazone
sulfa allergy
MOA of Allopurinol and Febuxostat
inhibits xanthine oxidase to decrease plasma urate levels
Side Effects of Allopurinol
Peripheral neuritis
Necrotizing vasculitis
aplastic anemia
bone marrow suppression
hypersensitivity
hepatitis
Reduce Azathioprine and 6MP
Side Effects of Febuxostat
Increased LFTs
Nausea
Arthralgias
Rash
MOA of Pegloticase
recombinant uricase that attaches to methoxy polyethylene glycol
