More easily identify cells, Lyson needed cells, look for specific things in cells
What stains well with Romanowski stain?
Organisms and cytoplasm, nuclear detail is usually sufficient
What does Romanowski poorly stain?
Mass cell granules, basofills, bone marrow cells
What is another name for Romanowski stain?
DiF quick
What is step one to DiF quick stain?
Fixative – 95% methanol, light blue
What is step 2 diff quick stain
Eosin g, acidic ph
What is step three to Diff quick stain?
New methylene blue
What is the pH of new methylene blue stain?
Alkaline

What are advantages to papanicolaou stains?
Excellent nuclear and cytoplasmic detail
What are disadvantages to papanicolaou stains
Multiple steps and time-consuming, harder to find, prepare, maintain, cells must be wet fixed, use a protein coated slide to prevent cells from lining off when immersed, can be expensive
What does it mean for a cell to be wet fixed?
Spray the smear with a cytologic fixative, or place ethanol immediately after prep
What stain should you use for an ear cytology?
Diff quick
What view should you view an ear cytology under?
Oil of immersion
