- put the tube containing cells in a hot water bath to melt cells/ unfreeze.
- put 1 μL of media
- put the tube in the centrifuge machine to separate cells from media. ( cells go down and media go on top ).
- remove the supernatants to remain with the cells at the bottom.
- Warm the medium to use in the culture.
- Add 1 μL of the medium again to the tube of cells and mix.
- put them in petri-dish.
- mix them well and put them in the incubator to grow.
- replace medium with the new one that has nutrients, along the growth period until you have the desired amount of cells to use.
