1. put the tube containing cells in a hot water bath to melt cells/ unfreeze.
2. put 1 μL of media
3. put the tube in the centrifuge machine to separate cells from media. ( cells go down and media go on top ).
4. remove the supernatants to remain with the cells at the bottom.
5. Warm the medium to use in the culture.
6. Add 1 μL of the medium again to the tube of cells and mix.
7. put them in petri-dish.
8. mix them well and put them in the incubator to grow.
9. replace medium with the new one that has nutrients, along the growth period until you have the desired amount of cells to use.
