1.3 Microscopy Flashcards
What is the function of the eye piece lens and objective lens?
- Magnify the object/ make the object look bigger
What is the function of the coarse focus and fine focus?
- Increase the clarity of the image/ make the image clearer
How do you calculate total magnification?
total magnification = eye piece lens X objective lens
What is magnification?
How much bigger the image is compared to the actual specimen
What is resolution?
The smallest distance between two points that can be seen as two points
How do you calculate image size?
image size = magnification X actual size of specimen
What is field of view?
The circular area you see in the light microscope
How do you calculate estimated size?
field of view (in mm) ÷ number of cells you can see across the field of view
Factors of a light microscope
- Works with light reflecting of an image
- Cheaper and more accessible than the electron microscope
- Allows animal, plant and bacteria cells to be compared and viewed
Factors of an electron microscope
- Works with electrons reflecting of an image
- Higher magnification (can see smaller objects)
- Higher resolution (can see more detail)
- Higher resolution means we can investigate more about the function of structures inside the cells (e.g. the way mitochondria provides energy for cells)
